Aunt Agatha!

(cover-dated February 1955; on-sale date of November 30, 1954 per the GCD)
Aunt Agatha (not sure if her last name would be Wayne or not) appeared about ten years before Dick Grayson's Aunt Harriet was introduced.
And believe it or not, Aunt Agatha did show up for a flashback in Grant Morrison's run (but not looking all that much like she did for her original appearance):
from Batman #656 (October 2006)
