Golden Age Manhunter (Donald "Dan" Richards)
First appearance of the Quality Comics character named "Manhunter" was in Police Comics #8, cover-dated March 1942 with an on-sale date of January 9, 1942 according to the GCD:
The Manhunter feature lasted in Police Comics until 1950, with the last episode in issue #101.
Oh, and when Roy Thomas decide to include many of the Quality Comics characters as originating on Earth-2 in All-Star Squadron, he did have the Dan Richards-version of Manhunter meet up with the Paul Kirk-version (published by DC in Adventure Comics around the same time) in All-Star Squadron #31 (March 1984):

I have loved Manhunter a lot, especially the Walt Simonson stories

According to the GCD, that cover was by Gill Fox.

And then there have been several other characters from DC who have since gone by the name "Manhunter", including Mark Shaw, Chase Lawler, and Kate Spencer.
